🔬Science and Technology Promotion Organization (JST): Development of an optical comb microscope capable of high-speed image acquisition

~ Anticipating live observation of transparent / nonfluorescent cells and application to quality inspection of micro semiconductor parts with nanometric irregularities ~

point

Since the image contrast of the confocal microscope is based on the light intensity, application to transparent and nonfluorescent cell samples and semiconductor samples with nanometric irregularities has been restricted.

Because the confocal microscope is based on point measurement, mechanical scanning (scanning) of the focal position is necessary for image acquisition, and it is difficult to acquire high-speed images.

By using optical comb and wavelength / space conversion, we have developed a method capable of collectively acquiring confocal microscopic images based on optical phase and light amplitude at high speed without scanning (scanless).

It is expected to be applied to time-series monitoring of three-dimensional cultured cells and detection of nanometer defects of micro semiconductor parts.
