We have developed a highly efficient synthetic method for “3-hydroxy-2-pyrazinecarboxamide,” which is a synthetic intermediate for favipiravir (avigan).

Development background:

This intermediate has been found to be effective as an antiviral agent against FMD.

It is a synthetic intermediate of favipiravir (trade name: Avigan), which has recently attracted worldwide attention.

Develop synthetic intermediates / new methods:

As an “key step to construct this intermediate / ring structure”, we have developed “an efficient method using a catalyst with water as a medium”.

With the current method, “for every 100 kilograms produced, more than the same amount of waste is generated.”

On the other hand, “The new method was able to reduce waste to almost zero.”

Future use:

The new method is an improvement within the current manufacturing route.

The group of presenters is also implementing improvements within other current manufacturing routes.

We are also improving the manufacturing route itself and developing a continuous flow manufacturing method.

Avigan Manufacturing:

In the future, it is expected that foreign countries will follow up on the manufacture of Avigan.

In order to secure the superiority of Avigan, “a manufacturing method that cannot be followed by other countries should be developed in Japan as soon as possible”.

In addition, it is desirable to establish a stable supply system for avigan in Japan.

To that end, “development of a continuous flow method that replaces the current batch method” is essential.

It is necessary to make full use of various science and technology toward the development of a series of manufacturing methods for Avegan.

It is not something that a company or a university can handle.

Under the initiative of the government, it is desirable to create and work on an all-Japan system in which industry, government and academia are united.
